The culture and proliferation of stem cells in the laboratory are prerequisites for their use in research or even cell therapy. The most common type of culture medium supplement widely used to provide the necessary growth factors for cell survival, adhesion, and growth is Fetal Bovine Serum (FBS). However, especially for therapeutic purposes, avoiding contamination with various animal pathogens (proteins, viruses, and bacteria) must be considered. In this regard, replacing Fetal Bovine Serum with Human Umbilical Cord Serum (HUCS) has currently attracted the attention of many researchers. Advantages of Human Umbilical Cord Blood Serum over Commercial Serums Available in the Market

The key advantage of using HUCS is that it can be easily and massively obtained from umbilical cord blood after childbirth without any ethical concerns. Furthermore, HUCS is free from animal-derived pathogens and has a much lower probability of causing an immune or allergic reaction.
HUCS contains rich sources of growth factors, neurotrophic factors, and cytokines with wound healing, anti-inflammatory, anti-aging, and anti-apoptotic properties. Also, compared to peripheral blood serum and FBS, it significantly contains higher concentrations of various biomolecules including interleukins 4, 5, 6, 7, 10, 15, MCP-1, SCF, SDF, as well as growth factors involved in tissue repair and regeneration processes such as EGF, TGF-α, TGF-β, FGF, PDGF, VEGF, and NGF. Therefore, it has a higher potential than FBS for culturing various human and animal cells and can be a good substitute for platelet lysate, FBS, or FCS in culturing various human cells.
It is noteworthy that, based on the results of studies conducted at Royan Stem Cell Technology Company, the efficiency of a culture medium containing 5% umbilical cord serum in culturing mesenchymal stem cells is equivalent to a culture medium containing 10% bovine serum (HyClone and Gibco). At a 5% umbilical cord serum concentration, not only does mesenchymal cell proliferation occur well, but their differentiation potential and surface marker expression are also well-preserved.
